Traditional antibody drug conjugants are conjured by lysine residues of antibodies or sulfhydryl groups obtained by reducing disulfide bonds of antibodies, but the conjugations have disadvantages of poor uniformity and low stability. A high degree of homogeneity of antibody drug conjugations can be obtained by site-specific coupling technology to ensure the reproducibility between batches in the production process. Several site-specific coupling techniques for antibody drug conjugations are reviewed.
